Immunoprecipitation:Sec8 AntibodyNBP3-29261] -Detection of human EXOC4/SEC8 by western blot of immunoprecipitates. Samples: Whole cell lysate (0.5 or 1.0 mg per IP reaction; 20% of IP loaded) from Jurkat cells prepared using NETN lysis buffer. Antibody: Affinity purified rabbit anti-EXOC4/SEC8 antibody NBP3-29261 (lot NBP3-29261-1) used for IP at 6 µg per reaction. EXOC4/SEC8 was also immunoprecipitated by rabbit anti-EXOC4/SEC8.. For blotting immunoprecipitated EXOC4/SEC8, A304-786A was used at 1 µg/ml. Detection: Chemiluminescence with an exposure time of 30 seconds.

Background: Sec8

Sec8 is encoded by this gene is a component of the exocyst complex, a multiple protein complex essential for targeting exocytic vesicles to specific docking sites on the plasma membrane. Though best characterized in yeast, the component proteins and functions of exocyst complex have been demonstrated to be highly conserved in higher eukaryotes. At least eight components of the exocyst complex, including this protein, are found to interact with the actin cytoskeletal remodeling and vesicle transport machinery. The complex is also essential for the biogenesis of epithelial cell surface polarity. Alternate transcriptional splice variants, encoding different isoforms, have been characterized.
