Immunoprecipitation:[NBP3-28988]-Detection of human SPTBN2/SCA5 by western blot of immunoprecipitates. Samples: Whole cell lysate from HeLa cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-SPTBN2/SCA5 antibody used for IP at 6 µg per reaction. SPTBN2/SCA5 was also immunoprecipitated by rabbit anti-SPTBN2/SCA5 antibody. For blotting immunoprecipitated SPTBN2/SCA5, was used at 0. 4 µg/ml. Detection: Chemiluminescence with an exposure time of 3 minutes.

Background: beta-III Spectrin
Mutations in this gene cause a form of spinocerebellar ataxia, SCA5, that is characterized by neurodegeneration, progressive locomotor incoordination, dysarthria, and uncoordinated eye movements. Mutations in spectrins are a previously unknown cause of ataxia and neurodegenerative disease that affect membrane proteins involved in glutamate signaling. Spectrin-beta IIIs have been recognized as ataxia disease genes and their mutations cause spinocerebellar ataxia type 5 (SCA5).
