Western Blot: uPAR Antibody (A6E8-R) [NBP3-33084] - Western blot analysis of uPAR on different lysates with Mouse anti-uPAR antibody (NBP3-33084) at 1/1,000 dilution.

Lane 1: U937 cell lysate
Lane 2: U937 treated with 200 nM PMA for 72 hours cell lysate

Lysates/proteins at 20 ug/Lane.

Predicted band size: 37 kDa
Observed band size: 37-60 kDa

Exposure time: 15 seconds;

4-20% SDS-PAGE gel.

Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (NBP3-33084) at 1/1,000 dilution was used in 5% NFDM/TBST at 4C overnight. Goat Anti-Mouse IgG - HRP Secondary Antibody at 1/50,000 dilution was used for 1 hour at room temperature.

Background: uPAR

uPAR (Urokinase plasminogen activator receptor) is a glycoprotein I anchored surface receptor specific for urokinase plasminogen activator (uPA). Upon binding to uPAR, uPA converts the surface bound, large serum beta globulin plasminogen to plasmin. Plasmin, which is also designated fibrinolysin, is a trypsin-like enzyme that acts on Arg-Lys bonds and induces pericellular proteolysis in fibrin and fibrinogen, and thereby contributes to the systematic activation of the coagulation cascade. uPA and uPAR are known to be overexpressed in mesenchymal and epithelial tumor cells and are required for tumor invasion and metastasis.
