Immunoprecipitation:WIPI2 AntibodyNBP3-29825] -Detection of human WIPI2 by western blot of immunoprecipitates. Samples: Whole cell lysate (0.5 or 1.0 mg per IP reaction; 20% of IP loaded) from HeLa cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-WIPI2 antibody NBP3-29825 (lot NBP3-29825-1) used for IP at 6 µg per reaction. WIPI2 was also immunoprecipitated by rabbit anti-WIPI2 antibody. For blotting immunoprecipitated WIPI2, NBP3-29825 was used at 0.4 µg/ml. Detection: Chemiluminescence with an exposure time of 10 seconds.

Background: WIPI2

WD40 repeat proteins are key components of many essential biologic functions. They regulate the assembly of multiprotein complexes by presenting a beta-propeller platform for simultaneous and reversible protein-protein interactions. Members of the WIPI subfamily of WD40 repeat proteins, such as WIPI2, have a 7-bladed propeller structure and contain a conserved motif for interaction with phospholipids (Proikas-Cezanne et al., 2004 [PubMed 15602573]).[supplied by OMIM]
