Product Description

CoraFluor™ 1, thiol reactive is a terbium-based time-resolved Förster resonance energy transfer (TR-FRET) donor for TR-FRET assay development, such as target engagement and ternary complex assays. This product is designed with maleimide functionality to conjugate to cysteines, making it suitable for labeling biomolecules, including antibodies, nanobodies, proteins and peptides. It is exceptionally bright, providing unmatched assay sensitivity and stability in biological media over other commercially available TR-FRET donors. CoraFluor™ 1, thiol reactive is compatible with common acceptor dyes such as FAM, TMR and Cyanine 5, SE (Cat. No. 5436), enabling seamless assay integration in high throughput screening applications.

    Make sure to reduce with TCEP, exchange or quench, bubble everything with inert gas; 2 fold molar excess works well.
    I used CoraFluor-1 to label recombinant human protein with single cysteine for a TR-FRET assay. Since the amount is small, I went with a 2-fold molar excess of the probe per cysteine. I reduced the protein with fresh TCEP, buffer exchanged, and bubbled everything with nitrogen to keep conditions reducing. Labeling worked great. The probe is super bright—I get ~700x signal over buffer blank at 490/10 nm emission using just 5 nM. Overall, really solid performance and great for TR-FRET, even with low amounts.
