Background: HLA B7

HLA B belongs to the HLA class I heavy chain paralogues. This class I molecule is a heterodimer consisting of a heavy chain and a light chain (beta 2 microglobulin). The heavy chain is anchored in the membrane. Class I molecules play a central role in the immune system by presenting peptides derived from the endoplasmic reticulum lumen. They are expressed in nearly all cells. The heavy chain is approximately 45 kDa. Hundreds of HLA B alleles have been described. Human Leukocyte Antigens are highly polymorphic proteins that are involved in the presentation of antigens to the T-cell receptor. There are two classes of HLA antigens, class I (HLA-A, HLA-B and HLA-C) and class II (HLA-D). This antibody can be used for HLA typing.

  • Q: We are interested in HLA-B7 antibodies that could be used for the immunohistochemistry of paraffin-embedded tissue. HLA B7 Antibody (BB7.1) offered in your company is described to fulfill those requirements (catalog # NB100-64159, 0.2mg). May I kindly ask you if you have a pictures of the performance of those antibodies for the IHC-P application? Unfortunately, we were quite unlucky with 3 HLA-B7 products and it seems to be not so easy to get a specific for HLA-B7 allele staining working for IHC-P. Therefore, we would like to ask if you can provide a test vial of NB100-64159? I haven't been testing BB7.1 clone of HLA-B7 antibodies from any other company. But I have the very last hope - may I ask you if you are planning to validate NB100-6415 BB7.1 clone for IHC-P? The only IHC-P applicable antibodies suggested by Biocompare are EP2624 antibodies from Abcam - those are unspecific (equally stain lymphocytes from HLA-B7pos and HLA-B7neg donors).

    A: I looked into this product for you and actually have found out it has not been tested in IHC, which is why we had no images listed. So far this product has only been validated for flow cytometry. Have you ever used this clone from another company? If so, this most likely will not work as well since it is a licensed clone. Unfortunately we are unable to provide any free samples at this time per company policy. Unfortunately we do not have anything else to provide for this target at this time as the other two antibodies are just the directly conjugated version of this clone. A good place to check would be Biocompare as this site compares various products from different biology companies and you may be able to find something there. Since NB100-64159 can stain in a Flow application there is a good possibility it should also be effective in IHC, but since we cannot guarantee this I can direct you to our Innovators Reward Program.
