40S Ribosomal Protein S6 (RPS6) is the major substrate of protein kinases, particularly p70 S6 kinase, in eukaryotic ribosomes. RPS6 phosphorylation at S235, S236, S240, and S244 upregulates the translation of mRNAs containing an oligopyrimidine tract at their transcriptional start sites. This phosphorylation is stimulated by growth factors, tumor promoting agents, and other mitogens. RPS6 is dephosphorylated during growth arrest.

Detection of Human Phospho-Ribosomal Protein S6/RPS6 (S235/S236) by Western Blot.
Western blot shows lysates of HEK293T human embryonic kidney cell line untreated (-) or treated (+) with 100 ng/mL Recombinant Human EGF (Catalog # 236-EG) for 30 minutes. PVDF membrane was probed with 1 µg/mL of Rat Anti-Human Phospho-Ribosomal Protein S6/RPS6 (S235/S236) Monoclonal Antibody (Catalog # MAB54361) followed by HRP-conjugated Anti-Rat IgG Secondary Antibody (Catalog # HAF005). A specific band was detected for Phospho-Ribosomal Protein S6/RPS6 at approximately 32 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.Phospho-Ribosomal Protein S6/RPS6 (S235/S236) in MCF‑7 Human Cell Line.
Ribosomal Protein S6/RPS6 was detected in immersion fixed MCF-7 human breast cancer cell line untreated or treated with Rapamycin using Rat Anti-Human Phospho-Ribosomal Protein S6/RPS6 (S235/S236) Monoclonal Antibody (Catalog # MAB54361) at 3 µg/mL for 3 hours at room temperature. Cells were stained using the NorthernLights™ 557-conjugated Anti-Rat IgG Secondary Antibody (red; Catalog # NL013) and counterstained with DAPI (blue). Specific staining was localized to cytoplasm.
