Flow Cytometry: OX40 Ligand/TNFSF4 Antibody (R4930 (Oxelumab)) - Chimeric [NBP2-52687] - Human peripheral blood leukocytes were fixed using 2% PFA, permeabilised using 0.5% Triton and stained with unimmunized rabbit IgG antibody (isotype control, black line) or the rabbit IgG-chimeric version of R4930 (NBP2-52687, blue line) at a dilution of 1:100 for 1h at RT. After washing, bound antibody was detected using a goat anti-rabbit IgG AlexaFluor 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.

Background: OX40 Ligand/TNFSF4

CD252 is a 35 kD member of the TNF superfamily also known as OX40L and CD134L. The OX40 ligand is expressed on activated B cells and antigen presenting cells. OX40 ligand interacts with OX40 antigen (CD134) expressed predominantly on activated T cells to increase proliferation and IL-2 production, and to enhance proliferation and immunoglobulin secretion in activated B cells. The RM134L antibody can inhibit OX40/OX40 ligand interactions and block the costimulatory activity of OX40L.
