Western Blot: AKAP12 Antibody [NBP2-66604] - Western blotting of total cellular protein from cultured rat aortic smooth muscle cells was prepared and analyzed. The protein was transferred to a PVDF membrane, blocked with 1X TBS, 0.1% Tween 20/ 5% NFDM, probed with 5 ug/mL of pre-immune serum 5 ug of AKAP12 antibody (a 3 second exposure is shown). The blot was then washed and probed with monoclonal anti-sheep IgG coupled to HRP at 1:10000, and detected with luminol.

Background: AKAP12

Activation of protein kinase C is a key signal transduction event in mesangial cell dedifferentiation and proliferation, yet little is known about downstream substrates or their roles in normal or diseased states. AKAP12 or SSeCKS, a novel protein kinase C substrate originally isolated as a src-suppressed negative mitogenic regulator in fibroblasts, controls actinbased cytoskeletal architecture and scaffolds key signaling kinases such as protein kinase C and protein kinase A. Activation of protein kinase C is a key signal transduction event in mesangial cell dedifferentiation. A role for AKAP12, a PKA/PKC scaffolding protein, has been implicated during the process of spermiogenesis and in the actin-based stellate morphology of mesangial cells.
