Total protein from mouse brain was separated on a 12% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with a 1:500 dilution of anti-CHT1 in 1% milk/TBST. Reactivity was detected with an anti-mouse HRP secondary antibody using chemiluminescence.

This CHT1 antibody is useful for Immunohistochemistry frozen sections and paraffin embedded sections and Western blot. In Western blot a band can be seen at ~65 kDa representing CHT and a band can also be seen at ~45 kDa after deglycosylation with PNGase F.

Background: CHT1

CHT1 (high affinity choline transporter 1) is a member of sodium:solute symporter (SSF) (TC 2.A.21) family that facilitates sodium/chloride-ion dependent import of choline from extracellular space to the neurons with high affinity. CHT1 is expressed in putamen, spinal cord and medulla, and is specific for cholinergic neurons. Acetyl choline is synthesized from choline and acetyl CoA by choline acetyltransferase in the cytoplasm of cholinergic neurons, and then incorporated into synaptic vesicles by VAChT (vesicular acetylcholine transporter). In the pre-synaptic terminals of cholinergic neurons, CHT1 mediates choline uptake which is the rate limiting step for acetyl-choline biosynthesis and is essential for post-natal viability. CHT1 been shown to be internalized in a clathrin dependent manner, whereas, the synaptic vesicles exocytosis enhances its cell surface expression; a mechanism that facilitates CHT1 functions at cellular surfaces with its predominant localization in the synaptic vesicles in cholinergic neurons. Moreover, Nedd4-2-mediated ubiquitination regulates CHT1 cell surface expression reflecting a possibility that treatments or drugs which inhibit CHT1-Nedd4-2 interactions might be useful for diseases involving decrease in acetylcholine level such as Alzheimer's disease. Moreover, polymorphic variation in CHT1 has been shown to predict early, subclinical measures of carotid atherosclerosis which may aid in understanding cholinergic-vagal processes potentially underlying atherosclerotic risk.

Western Blot Protocol

1. Perform SDS-PAGE on samples to be analyzed, loading ~30 ug of total protein per lane.
2. Transfer proteins to PVDF according to the instructions provided by the manufacturer of the transfer
apparatus.
3. Rinse membrane with dH2O and then stain the blot using Ponceau S for 1-2 minutes to access the transfer of proteins onto the nitrocellulose membrane. Rinse the blot in water to remove excess stain and mark the lane locations and locations of molecular weight markers using a pencil.
4. Rinse the blot in PBS for approximately 5 minutes.
5. Block the membrane using 5% NFDM in PBS + Tween, 1 hour at RT.
6. Rinse the membrane in dH2O and then wash the membrane in wash buffer [PBS + 0.5% Tween] 3 times for 10 minutes each.
7. Dilute the mouse anti-hCHT primary antibody (NB 110-74570) in blocking buffer and incubate 1 hour at room temperature.
8. Rinse the membrane in dH2O and then wash the membrane in wash buffer [PBS + 0.5% Tween] 3 times for 10 minutes each.
9. Apply the diluted mouse-IgG HRP-conjugated secondary antibody in blocking buffer (as per manufacturers
instructions) and incubate 1 hour at room temperature.
10. Wash the blot in wash buffer [PBS + 0.5% Tween] 3 times for 10 minutes each (this step can be repeated as required to reduce background).
11. Apply the detection reagent of choice in accordance with the manufacturers instructions (Amersham ECL Plus).

Note: Tween-20 can be added to the blocking or antibody dilution buffer at a final concentration of 0.05-0.2%, provided it does not interfere with antibody-antigen binding.
