Phospho-Crk (Tyr-207) synthetic peptide (coupled to KLH) corresponding to amino acid residues surrounding Tyr-207 in human CrkL. This peptide sequence is well conserved in mouse and rat CrkL, and has 40% homology with the conserved site in Crk II (Tyr-221). The site is not found in Crk I. [UniProt# P46109]

The blot was treated with alkaline phosphatase to dephosphorylate CrkL (lanes 2 & 4), then the blot was probed with rabbit polyclonals CrkL (C-terminus) NBP3-23256 (lanes 1 & 2) and CrkL (Tyr-207) phospho-specific NBP3-23257 (lanes 3 & 4).

Background: CrkL

CrkL is a widely-expressed 39 kD adaptor protein that belongs to the Crk protein family and is abundantly expressed in cells of hematopoetic origin. Through its SH2 and SH3 domains CrkL initiates interactions with numerous downstream proteins such as CBL, Cas, Paxillin, C3G, SOS, PI3K, c-Abl and BCR/Abl. It is involved in processes such as adhesion, migration and immune responses. Furthermore, CrkL plays an essential role in Chronic Myelogenous Leukemia and is constitutively phosphorylated by the Bcr-Abl kinase in this disease.
