Western Blot: EGLN3/PHD3 Antibody (JE60-82) [NBP3-32745] - Western blot analysis of EGLN3/PHD3 on different lysates with Rabbit anti-EGLN3/PHD3 antibody (NBP3-32745) at 1/1,000 dilution.

4-20% SDS-PAGE gel.

Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (NBP3-32745) at 1/1,000 dilution was used in 5% NFDM/TBST at 4 overnight. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1/50,000 dilution was used for 1 hour at room temperature.

Background: EGLN3/PHD3

HIF prolyl hydroxylase 3 (HPH-3; PHD3; EGLN3) catalyzes the post-translational formation of 4-hydroxyproline in hypoxia-inducible factor (HIF) alpha proteins, hydroxylates HIF-1 alpha at Pro-564, and also hydroxylates HIF-2 alpha. PHD3 functions as a cellular oxygen sensor and, under normoxic conditions, targets HIF through the hydroxylation for proteasomal degradation via the von Hippel-Lindau ubiquitylation complex. EGLN3 may play a role in cell growth regulation in muscle cells and in apoptosis in neuronal tissue. HIF prolyl hydroxylase 3 promotes cell death through a caspase-dependent mechanism.
