Flow Cytometry: Galectin-9 Antibody (RG9-35) - Chimeric - Azide and BSA Free [NBP3-09032] - NIH3T3 cells were fixed using 2% PFA and stained with anti-unknown specificity antibody (isotype control, black line) or NBP3-09032 (blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.

Immunocytochemistry /Immunofluorescence: Galectin-9 Antibody (RG9-35) - Chimeric - Azide and BSA Free [NBP3-09032] - Immunofluorescence analysis of paraformaldehyde fixed RAW264.7 cells on Shi-fix(TM) coverslips stained with NBP3-09032 at 10 ug/ml for 1h followed by Alexa Fluor(R) 488 secondary antibody (2 ug/ml), showing membrane staining. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom: NBP3-09032, DAPI, merged channels and an isotype control. The isotype control was an unknown specificity antibody followed by staining with Alexa Fluor(R) 488 secondary antibody.

Background: Galectin-9

The galectins are a family of beta-galactoside-binding proteins implicated in modulating cell-cell and cell-matrix interactions. Galectin 9 is an S-type lectin and an eosinophil chemoattractant (ECA) produced by antigen-activated T cells. Galectin 9 is involved in apoptosis, is thought to be a prognostic factor for breast cancer and is also overexpressed in Hodgkin's disease tissue. The protein has N- and C- terminal carbohydrate-binding domains connected by a link peptide. Multiple alternatively spliced transcript variants have been found for this gene.
