Flow Cytometry: GR/NR3C1 Antibody (BuGR2) [PE] [NB300-731PE] - An intracellular stain was performed on Jurkat cells with GR/NR3C1 (BuGR2) antibody NB300-731PE (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 5 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to Phycoerthrin.

Background: GR/NR3C1

Steroid receptors are ligand-dependent, intracellular proteins that stimulate transcription of specific genes by binding to specific DNA sequences following activation by the appropriate hormone. Glucocorticoids are a family of steroids necessary for the regulation of energy metabolism and the immune and inflammatory responses. These compounds exert their effect through their interaction with the glucocoticoid receptor (GR) and that complex's subsequent association with DNA. All normal mammalian tissues examined to date have been shown to contain glucocorticoid receptor. The corresponding gene for the glucocorticoid receptor is NR3C1.

  • Q: I'm wondering if you can tell me more about your post-conjugation Q/C process. What kind of Q/C tests do you run, and how do you determine whether the conjugation is good or bad (or said another way, passes or fails)?

    A: We QC test every newly produced lot of antibody (unconjugated). Once the unconjugated product meets our testing standards, we release that antibody for sale. When it comes to the conjugations, we are not able to individually test every single conjugation in each application, as these are made-to-order (we only conjugate what is needed to fulfill customer orders). For PE specifically, we perform a 3:1 ratio of dye to protein, purify via HLPC, calculate the F/P ratio, and concentrate/calculate end concentration. As long as the conjugated antibody meets our F/P ratio standards and concentration standards, we release it for shipment.
