Q: A while back I ordered around 18 antibodies and have been in the process of testing them. And I realized that a couple of them under the application section on the website said: Antibody reactive against Recombinant Protein with GST tag on ELISA and Western Blot and also on transfected lysate in western blot. GST tag alone is used as a negative control. I was wondering if that means the antibody won't detect the endogenous epitope and will only detect the recombinant protein that's been transfected? Or if it can be used also to detect the endogenously expressed protein. So the two antibodies in question are the H00050810-D01P Hdgfrp3 antibody and the H00000440-D01P Asparagine synthetase antibody. Both show a transfected lysate as a positive control for the antibodies. Are you aware of any positive control cell lines for detecting endogenous (non-transfected) protein for either or both of these antibodies?
A: The antibodies you described should be able to detect the endogenous protein, and we guarantee them for this purpose. We do our best to provide as much information as possible about the testing conditions of our antibodies, which is why we included the stated information. I do not know of any non-transfected cell lines that are suitable as specific positive controls for either asparagine synthetase or Hdgfrp3. K563 cells and muscle tissue express asparagine synthetase, but this could be influenced by treatment of the cells. MCF-7 cells may be a good positive control for Hdgfrp3.
