Background: AIM2
Human Interferon-inducible protein AIM2 (also known as absent in melanoma 2 or simply AIM2), encoded by the AIM2 gene, is a 343 amino acids (aa), 39 KDa protein with a N-terminal DAPIN (or pyrin) domain (amino acids 1-87) and a C-terminal HIN-200 domain (amino acids 138-337), which is known to have two oligonucleotide-binding folds. Recent research has shown that AIM2 is part of the inflammasome and is involved in innate immune response by recognizing cytosolic double-stranded DNA and inducing caspase-1-activating the inflammasome in macrophages. Upon binding to DNA, Aim-2 undergoes oligomerization and associates with PYCARD initiating the recruitment of caspase-1 precursors and processing of IL-1b and IL-18. Over aa 71-180, human Aim2 shares 90% aa sequence identity with macaque Aim2.
