Detection of UVRAG in Daudi Human Cell Line by Flow Cytometry. Daudi human Burkitt's lymphoma cell line was stained with Mouse Anti-Human UVRAG Alexa Fluor® 647‑conjugated Monoclonal Antibody (Catalog # IC9180R, filled histogram) or isotype control antibody (Catalog # IC002R, open histogram). To facilitate intracellular staining, cells were fixed with Flow Cytometry Fixation Buffer (Catalog # FC004) and permeabilized with Flow Cytometry Permeabilization/Wash Buffer I (Catalog # FC005). UV radiation resistance-associated gene protein, or UVRAG, is a multi-function protein of 699 amino acids involved in the regulation of cellular pathways implicated in membrane trafficking. UVRAG activates the Beclin-1-PI3KC3 complex, a critical element in the autophagy signaling pathway. Binding studies show that UVRAG and Beclin-1 directly interact via their coiled-coil domains and that Beclin-1 functions as a platform for the formation of the Bcl-2-UVRAG-PI3KC3 complex. Chromosomal aberrations involving the UVRAG gene are associated with left-right axis malformation, and mutations in the gene have been associated with colon cancer.
