Background: IBSP/Sialoprotein II

The protein encoded by the IBSP gene is a major structural protein of the bone matrix. It constitutes approximately 12% of the noncollagenous proteins in human bone and is synthesized by skeletal-associated cell types, including hypertrophic chondrocytes, osteoblasts, osteocytes, and osteoclasts. The only extraskeletal site of its synthesis is the trophoblast. This protein binds to calcium and hydroxyapatite via its acidic amino acid clusters, and mediates cell attachment through an RGD sequence that recognizes the vitronectin receptor.

    This is the image of BSPII staining of 14 day old mouse proximal tibia. BSPII is detected by green secondary antibody, however, the stain did not work. Likely due to the fact that this antibody can only be used in humans and possibly only for WB.
    Our previous studies have shown that BSPII has high expression in the epiphysis of the developing mouse proximal tibia and distal femur. To visualize this, we stained the tissue with BSPII antibody (NBP1-70422). As it was tested to work in human tissue and with western blots and the fact that there were not many other antibody options with Novus, we attempted the same with mouse tissue. We tested both 1:50 and 1:100 concentrations due to the higher concentrations typically needed when performing IHC vs Western blot. Unfortunately our stains came up negative/unspecific. This data was controlled for by another BSPII antibody we obtained from another vendor years ago.
