Application: ImmunocytochemistrySample Tested: Mouse ESC derived cells and differentiated ESC culturesSpecies: MouseVerified Customer | Posted 08/27/2021This antibody seems to not recognize Mesp2 in IF, as we see no nuclear staining using a protocol inducing mesodermal fate (S Morgani et al., 2018; JL Plouhinec et al., 2020). The expression is similar to a negative control. The protocol used is the following : Fixation for 30 min in PFA, 4%, RT 3 washes for 5 min in PBS, RT Permeabilization and blocking for 1 hour in PBS + 3% BSA + 0.1% Triton, 4°C Primary antibody reaction OverNight in PBS + 3% BSA + 0.1% Triton + primary AB, 4°C (Mesp2 at 1ug/ml) 3 washes for 5 min in PBS, RT Blocking for 1 hour in PBS + 3% BSA + 0.1% Triton, 4°C Secondary antibody reaction for 2h in PBS + 3% BSA + 0.1% Triton + 1/500 secondary AB (Dylight 650 Donkey anti Rabbit), 20°C 3 washes for 15 min in PBS, RT (Second wash with 5um Hoechst)Bio-Techne ResponseThis review was submitted through the legacy Novus Innovators Program, reflecting a new species or application tested on a primary antibody.
