Background: MESP2

MESP2 is a gene that codes for a protein that is 397 amino acids long, weighs approximately 42 kDa, and functions as a transcription factor that plays a role in somitogenesis and the development of cardiac mesoderm as well as regulates the FGF signaling pathway. Current research is being done on diseases and disorders related to this gene including dysostosis, alagille syndrome, cadasil, and scoliosis.

    This antibody seems to not recognize Mesp2 in IF, as we see no nuclear staining using a protocol inducing mesodermal fate (S Morgani et al., 2018; JL Plouhinec et al., 2020). The expression is similar to a negative control.
    The protocol used is the following : Fixation for 30 min in PFA, 4%, RT 3 washes for 5 min in PBS, RT Permeabilization and blocking for 1 hour in PBS + 3% BSA + 0.1% Triton, 4°C Primary antibody reaction OverNight in PBS + 3% BSA + 0.1% Triton + primary AB, 4°C (Mesp2 at 1ug/ml) 3 washes for 5 min in PBS, RT Blocking for 1 hour in PBS + 3% BSA + 0.1% Triton, 4°C Secondary antibody reaction for 2h in PBS + 3% BSA + 0.1% Triton + 1/500 secondary AB (Dylight 650 Donkey anti Rabbit), 20°C 3 washes for 15 min in PBS, RT (Second wash with 5um Hoechst)
