This A4 antibody has been tested by flow cytometric analysis of 3-day PHA-activated PBMC. This can be used at less than or equal to 0.5 ug per test. Cell number should be determined empirically but can range from 10^5to 10^8cells/test.

Background: MHC Class I

The A4 monoclonal antibody reacts with the class I human leukocyte antigen (HLA) complex in the absence of beta2 microglobulin. There are three class I alpha-chain genes in humans, called HLA-A, -B and -C. Normally, HLA class I proteins are associated non-covalently with beta2 microglobulin (beta2M). The function of the HLA class I complex is to present peptides derived from intracellular pathogens, including viral pathogens, on the cell surface to antigen specific CD8+ cytotoxic T cells. Class I HLA is expressed by the majority of nucleated cells.
