Background: MAGP-1/MFAP2
MAGP-1 (Microfibril-associated glycoprotein 1; also MFAP-2) is a 30‑33 kDa member of the MFAP family of proteins. It is secreted by endothelial and smooth muscle cells. MAGP-1 apparently mediates the release of Notch extracellular domain and also likely binds to type IV collagen and contributes to elastin fiber formation. Mature mouse MAGP-1 is 167 amino acids (aa) in length. It contains a transglutamination site at Gln20, a tyrosine sulfation motif (aa 46‑49), two potential O-linked glycosylation sites (Thr56 and Thr76), and a Cys-rich C-terminus that generates intermolecular disulfide bonds. One potential splice variant exists that is strictly intracellular and a second shows a 22 aa substitution for the C-terminal 127 amino acids. Mature mouse MAGP-1 is 94% and 87% aa identical to rat and human MAGP‑1, respectively.
