Application: Immunohistochemistry-Free Floating sectionsSample Tested: Mouse Brain TissueSpecies: MouseVerified Customer | Posted 12/05/2016The laser settings were exactly the same for both the CHRNA7 image and the no primary control. For the purpose of comparison, no image adjustments were made - these are jpegs from the raw.nd2 file z stack (1um steps).Tissue Mouse brain tissue was perfused with 4% Paraformaldehyde in phosphate buffered saline (1x PBS) Brains were immersed in 30% sucrose in 1x PBS and sectioned at 40um on a microtome Slices were stored in cryoprotectant solution at -20C until used for immunohistochemistry Immunohistochemistry Free floating sections were blocked with 5% normal goat serum (NGS) in 0.3% triton in 1x PBS The primary antibody, rb anti CHRNA7 (NBP1-79948), was used at 1:500 and incubated for two hours at room temperature followed by 12 hours at 4C. As a negative control, slices were incubated with blocking buffer and no primary antibody. The secondary antibody, Goat anti rabbit 633 (Alexa fluor), was used at 1:2000 and incubated with the slices for 90 minutes. Slices were mounted on glass slides with mounting media containing DAPI. Results CHRNA7 (white) with DAPI (blue) in the CA1 region of the hippocampus. My conclusion is that the CHRNA7 antibody does work in IHC. Images were taken using a Nikon confocal microscope and a 60x objective with 1.5 N.A.. The images are 1024 x 1024 pixels. The laser settings were exactly the same for both the CHRNA7 image and the no primary control. For the purpose of comparison, no image adjustments were made - these are jpegs from the raw.nd2 file z stack (1um steps).
