Immunocytochemistry/Immunofluorescence: non-muscle Myosin IIA Antibody (2B3) [H00004627-M03] - Bovine ovarian granulosa cells. Anti Myosin IIA immunofluorescent staining (green). DNA in counterstained with DAPI (blue). Control was obtained by omitting the primary antibody. Fixation: 4% PF for 7 minutes at rt. Permeabilization: 0.1% Triton-X in PBS for 7 minutes. Blocking: 20% donkey serum, 1% BSA in PBS, 1 hour at rt. Primary antibody: 1:50 or 1:100 in 1% BSA/PBS over night at 4C. Secondary antibody: donkey anti mouse -Alexa 488 (Invitrogen), 1:500. DNA Counterstaining: mounting medium with DAPI. Negative CTRL was performed by omitting the primary antibody, images were captured under an epifluorescence microscope.
