Antibodies: Affinity purified rabbit anti-REST antibody NB100-756 used for IP at 6 ug per reaction. For blotting immunoprecipitated REST, NB100-756 was used at 1 ug/ml. Detection: Chemiluminescence with an exposure time of 3 minutes.

Background: REST/NRSF

REST, which is short for RE1-silencing transcription factor, locks down neuronal genes in other cells by grabbing onto the DNA and cementing in other molecules, an arrangement that stays intact as non-neuronal cells differentiate into liver, muscle, and other tissues. REST uses a different temporary off mechanism to direct neuronal development. REST is a tumor suppressor gene in epithelial cells and has been found to be mutated in 33 percent of colorectal tumors and cell lines studied. Rest could eventually serve as a marker for diagnosing colorectal and other types of cancer.

  • Q: The customer is complaining that the antibody NB100-756 could not detect the target band and there is non-specific band in western blotting. Much detailed result is enclosed with the mail, please check it and give us some help.

    A: Your customer is observing a band at the same size that we observe for NRSF. The molecular weight of larger proteins can be harder to estimate due to poor resolution afforded by molecular weight ladders, and to the greater degree of variability in their migration through SDS-PAGE gels. NRSF is also expressed as a number of different isoforms, which is more than likely what your customer is detecting.
