Specificity
NB100-63208 recognizes the C-terminal of human pericentrin 1, a 75 kDa component of the filamentous matrix of the centrosome, expressed in all tissues. When associated with centrosome and Golgi localized protein kinase N-associated protein (CG-NAP), pericentrin 1 anchors the gamma-tubulin ring complex, providing a site for microtubule nucleation and the initial establishment of organized microtubule arrays. It binds calmodulin and is a constituent of nuclear pore proteins (nup) 107-160, a sub-complex of the nuclear pore complex. Pericentrin 1 is thought to play a role in the functioning of the centrosomes, cytoskeleton, and cell-cycle progression.

Application: ImmunofluorescenceSample Tested: RPE1 cellsSpecies: HumanVerified Customer | Posted 07/26/2021RPE1-hTERT cells are stained for pericentrin (Red) and primary cilia (Green). Pericentrin should be localized on the centrosome, base of the primary cilia. Based on the staining, there is no specific localization on the centrosome.IF done on 4% PFA (10min) and methanol (5min) fixed RPE1-hTERT cells. blocked with 5% BSA for 1hr at room temp. Primary antibodies: goat anti-pericentrin (1:100) and rabbit anti-Arl13B (1:500). Secondary antibodies: donkey anti-goat alexa fluor 555 and donkey anti-rabit alexa fluor 488.Bio-Techne ResponseThis review was submitted through the legacy Novus Innovators Program, reflecting a new species or application tested on a primary antibody.
