Phospho-Profilin (Tyr-129) synthetic peptide (coupled to carrier protein) that includes amino acids surrounding Tyr-129 in human Profilin 1. This sequence is well conserved in rat and mouse Profilin 1. The tyrosine site has some homology to the conserved site (Tyr-129) in Profilin-2a and Profilin-2b, but the site is not conserved in Profiin-3 and Profilin-4. [UniProt# P07737]

Specificity
This antibody was cross-adsorbed to unphosphorylated Profilin 1 (Tyr-129) then affinity purified using Profilin 1 (Tyr-129) peptide (without carrier). The antibody detects a 15 kDa band corresponding to the apparent molecular mass of Profilin 1 in human umbilical vein endothelial cells (HUVEC) and jurkat cells treated with pervanadate. This reactivity is removed by alkaline phosphatase treatment.

Western Blot: Profilin 1 [p Tyr129] Antibody [NBP3-23345] - Western blot of HUVEC stimulated with Pervanadate (1 mM) for 30 min (lanes 1-4). The blots were treated with alkaline phosphatase to remove phosphorylation (lanes 2 & 4), then probed with anti-Profilin (a.a. 126-137) (lanes 1 & 2) or anti-Profilin (Tyr-129) phospho-specific (lanes 3 & 4). The antibodies detect profilin at 15 kDa. In lanes 3 & 4, the antibody also detects an unknown 20 kDa protein.Applications for Profilin 1 [p Tyr129] Antibody
