Chromatin Immunoprecipitation: RUNX1T1/ETO Antibody [NBP2-59232] - ChIP assays were performed using SKNO-1 cells, the antibody against ETO and optimized primer pairs for qPCR. Sheared chromatin from 1.25 million cells and 4 uL of antibody were used per ChIP experiment. QPCR was performed using primers specific for the FUT7, NFE2, OGG1 and VEGF genes. Figure shows the occupancy, calculated as the ratio + control/background for which the H2B gene was used.

ELISA: RUNX1T1/ETO Antibody [NBP2-59232] - To determine the titer of the antibody, an ELISA was performed using a serial dilution of the antibody directed against human ETO. The plates were coated with the peptides used for immunization of the rabbit. By plotting the absorbance against the antibody dilution, the titer of the antibody was estimated to be 1:1,300.

Background: RUNX1T1/ETO

RUNX1T1 / ETO is encoded by this gene is a putative zinc finger transcription factor and oncoprotein. In acute myeloid leukemia, especially in the M2 subtype, the t(8;21)(q22;q22) translocation is one of the most frequent karyotypic abnormalities. The translocation produces a chimeric gene made up of the 5'-region of the RUNX1 gene fused to the 3'-region of this gene. The chimeric protein is thought to associate with the nuclear corepressor/histone deacetylase complex to block hematopoietic differentiation. Several transcript variants encoding multiple isoforms have been found for this gene.
