The mouse monoclonal antibody S-HCL-1 (also known as S-HCL1) recognizes an extracellular epitope of CD22 (BL-CAM), a 130 kDa type I transmembrane glycoprotein (immunoglobulin superfamily) expressed in the cytoplasm of pro-B and pre-B lymphocytes, and on the surface of mature and activated B lymphocytes; it is lost on plasma cells, peripheral blood T lymphocytes, granulocytes and monocytes.

Western Blot: Mouse Monoclonal Siglec-2/CD22 Antibody (S-HCL-1) [NBP3-09167] - The reactivity of S-HCL-1 antibody was assessed by comparing binding signals in a panel of three human B cell and one T cell line. Western blotting analysis was performed on whole cell extracts (RIPA lysis buffer) of Raji, Ramos, Daudi, and Jurkat cell lines, mixed and heated (100C, 5 min) without reducing agent in SDS-loading buffer. Samples were resolved using 7% SDS-PAGE gel.Nitrocellulose membrane blot was probed with mouse IgG2b monoclonal antibody S-HCL-1 (2 ug/ml), followed by IRDye 800CW Goat-anti-Mouse IgG (green).CD22 was detected at ~110-125 kDa in Daudi cell line.

Background: Siglec-2/CD22

CD22 is a single-pass type 1 membrane glycoprotein found in the cytoplasm and cell membrane of B-lymphocytes. CD22 has at least two isoforms (alpha and beta) produced by alternate splicing. The protein exists predominantly as a monomer of the beta isoform (95 kDa). It is also found as a heterodimer of the beta and alpha isoforms.
