Western Blot:[NBP3-30449]-Detection of human and mouse SPTLC1 by western blot and immunoprecipitation. Samples: Whole cell lysate from HeLa, HEK293T, Jurkat and mouse NIH 3T3 cells. Antibodies: Affinity purified rabbit anti-SPTLC1 antibody used for WB at 0. 04 µg/ml and 0. 4 µg/ml and used for IP at 6 µg/mg lysate. SPTLC1 was also immunoprecipitated by rabbit anti-SPTLC1 antibody, which recognizes a downstream epitope. Detection: Chemiluminescence with exposure times of 30 seconds and 10 seconds.

Background: SPTLC1

Serine palmitoyltransferase, which consists of two different subunits, is the key enzyme in sphingolipid biosynthesis. It converts L-serine and palmitoyl-CoA to 3-oxosphinganine with pyridoxal 5'-phosphate as a cofactor. The product of this gene is the long chain base subunit 1 of serine palmitoyltransferase. Mutations in this gene were identified in patients with hereditary sensory neuropathy type 1. Alternatively spliced variants encoding different isoforms have been identified. (provided by RefSeq)
