Background: Thyroxine

This assay employs the competitive inhibition enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with an antibody specific to thyroxine. Standards or samples are added to the appropriate microtiter plate wells with Biotin-conjugated thyroxine. A competitive inhibition reaction is launched between thyroxine (Standards or samples) and Biotin-conjugated thyroxine with the pre-coated antibody specific for thyroxine. The more amount of thyroxine in samples, the less antibody bound by Biotin-conjugated thyroxine. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Substrate solution is added to the wells and the color develops in opposite to the amount of thyroxine in the sample. The color development is stopped and the intensity of the color is measured.
