CB2 and TRPV1 expression in OCs derived from CCS.Protein expression of CB2 (A) and TRPV1 (B) in OCs from 5 Childhood cancer survivors (CCS) compared with OCs from 5 healthy subjects (CTR), determined by Western Blot starting from 15μg of total lysate. The most representative images are showed. The Image Studio Digits software has been used for the protein bands density detection. The intensity ratios of immunoblots compared to CTR, considered as 1, were quantified after normalization with the housekeeping protein beta -Actin. Histogram shows the relative quantification for CB2 and TRPV1 expression as mean +/- SD of independent experiments on each individual sample. For statistical analysis it has been used a t-test. *Indicates p ≤ 0.05 compared to CTR. In Western Blot, a band is seen at ~100 kDa representing TRPV1. The observed molecular weight of the protein may vary from the listed predicted molecular weight due to post translational modifications, post translation cleavages, relative charges, and other experimental factors. In ICC/IF, plasma membrane staining is observed in Ntera-2 cells. This antibody is not recommended for IHC-paraffin embedded sections.

Background: Vanilloid R1/TRPV1

TRPV1 (transient receptor potential cation channel subfamily V member 1, capsaicin receptor, vanilloid receptor, Vr1) is a polymodal nociceptor activated by heat, vanilloids (capsaicin), and protons. It is primarily expressed in the dorsal root ganglia and peripheral nerve endings. TRPV1 antagonists are a potential therapeutic target for many suffering chronic physical and neuropathic pain. However, due to its other role in the regulation of body temperature, these therapies are currently experiencing a barrier. On the other hand, TRPV1 agonists can be used to desensitize nerves after activation, thereby reducing pain. The research around TRPV1 is broad, where it is studied in fields such as anxiety, pain, oxidative stress, taste, inflammation, depression, respiratory disease, and various psychiatric disorders.
