Immunoprecipitation: XPV/DNA polymerase eta Antibody [NB100-60424] - Detection of human XPV/DNA polymerase eta/PolH by western blot of immunoprecipitates. Samples: Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from Hela cells prepared using NETN lysis buffer. Antibodies: Affinity purified Rabbit anti-PolH antibody NB100-60424 (lot 3) used for IP at 6 ug per reaction. PolH was also immunoprecipitated by a previous lot of this antibody (lot 2) and rabbit anti-PolH antibody NB100-60423. For blotting immunoprecipitated PolH, NB100-60424 was used at 0.1 ug/ml. Chemiluminescence with an exposure time of 3 minutes.

Background: XPV

DNA polymerase eta is specifically involved in DNA repair. It plays an important role in translesion synthesis, where the normal high fidelity DNA polymerases cannot proceed and DNA synthesis stalls. It also plays an important role in the repair of UV-induced pyrimidine dimers. Depending on the context, it inserts the correct base, but causes frequent base transitions and transversions. It targets POLI to replication foci (referenced from swissprot).
