Background: ApoE4

Apolipoprotein E has three isoforms namely ApoE2, ApoE3, and ApoE4 that differ from each other by single amino acid interchanges at residues 130/176, and ApoE4 is an isoforms which contains arginine at 130 and 176 locations. ApoE4 is mainly synthesized in liver and brain, and is widely expressed in other tissues also. ApoE4 functions have been linked to neuronal plasticity, synaptogenesis etc. and ApoE4 expressing subjects have reduced capacity for synaptic plasticity, increased amyloid-beta deposition, and early onset of Alzheimer's disease (AD). ApoE4 is also linked to progression/poor clinical outcomes in TBI (traumatic brain injury), multiple sclerosis, stroke, frontotemporal dementia, and Parkinson's disease. The neuropathological effects of apoE4 includes impaired neurite outgrowth, cytoskeletal disruption/hyperphosphorylation of tau, neuronal mitochondrial dysfunction, impaired synaptogenesis, increased amyloid beta production as well as clearance/deposition, increased lysosomal leakage and neuronal apoptosis, cerebral neuropathology and impaired learning and memory.
