Effects of siRNA-mediated knockdown of expression of AldoA, EWSR1 and ILF3-90 on enterovirus replication.A, B. HeLa cells were transfected with siRNAs specific to AldoA, EWSR1 and 90KDa isoform of ILF3, or non-targeting control siRNA, and polio or Coxsakie B3 replicon replication assays were performed 72 h post siRNA transfection. The total replication signal was calculated as the area under the corresponding kinetics curves. Cell viability signal is proportional to the level of ATP in cells. Western blots show the efficacy of siRNA-mediated knockdown of the targeted proteins. C. HeLa cells were transfected with siRNAs specific to AldoA, EWSR1 and 90KDa isoform of ILF3, or non-targeting control siRNA. 72 h post siRNA transfection cells were infected with an MOI of 1 PFU/cell of poliovirus or Coxsackie virus B3, and the total virus yield was determined at 6 h p. i. Western blots show the efficacy of siRNA-mediated knockdown of the targeted proteins. D. HeLa cells were transfected with siRNAs specific to 90KDa isoform of ILF3 or EWSR1 or a non-targeting control siRNA. 72h post siRNA transfection, the cells were transfected with a replication-defective replicon RNA containing the delta 3D mutation. Background: EWSR1

EWSR1 (Ewing sarcoma breakpoint region 1) belongs to TET family (Tls, EWSR1, TafII) of RNA-binding proteins and it is implicated in transcriptional regulation for specific genes, mRNA splicing and transcription initiation. EWSR1 has the potential to associate with basal transcription factor TFIID and RNA polymerase II complex, and is required for proper localization of aurora B during mitosis as well as for maintaining mitotic spindle integrity. EWSR1 also associates with EP300/CREBBP as coactivator and EWSR1-EP300/CREBBP facilitates FOS activation as well as HNF4 genes activation. EWSR1 activates other transcription factors such as POU4F1 (or BRN3A, 13q13) and POU5F1 (or OCT4, 6p21) which regulate differentiation of neuronal cells. EWSR1 and CCNL1 are also interacting partners of TFIP11 which is functionally related to spliceosome and involved in pre-mRNA splicing. EWSR1 is required for cell survival in CNS and performs important functions in regulating genomic integrity and in RNA/microRNA's maturation processes. EWSR1 is ubiquity expressed with dominant localization in the nucleus and rarely in cytoplasm or various subcellular compartments, depending on the methylation state of its RNA-binding domain. Several gene families, mainly encoding for transcriptional regulators, can translocate with EWSR1 and the chimeric proteins have an aberrant activity interfering with different molecular pathways crucial for cell growth, differentiation and proliferation. EWSR1 is involved in several sarcoma translocations and soft tissue myoepithelial tumors.
