Immunogen

This antibody was raised by immunising C57Bl/6 mice with IL-17/IL-17A cross-linked to ovalbumin to generate a highly immunogenic complex.

Specificity

This antibody is highly specific for murine IL-17/IL-17A; in ELISA analysis, no cross-reactivity with IL-17B, IL-17C, IL-17D, IL-17E or IL-17F was observed.

Flow Cytometry: IL-17/IL-17A Antibody (MM17F3) - Chimeric [NBP3-12035] - Mouse splenocytes were stimulated with PMA and Ionomycin in presence of Monensin for 4 hours. The cells were fixed using 2% PFA, permeabilized using 0.5% Triton and stained with anti-Fluorescein IgG antibody (4-4-20; isotype control, black line) or the rabbit IgG version of MM17F3 (NBP3-12035, blue line) at a dilution of 1:100 for 1h at RT. After washing, bound antibody was detected using a goat anti-mouse IgG AlexaFluor(R) 488 antibody at a dilution of 1:1000 and cells were analyzed using a FACSCanto flow-cytometer.

Background: IL-17/IL-17A

Interleukin 17 (IL-17) is a pro-inflammatory cytokine produced by TCRalpha/beta+ CD4 CD8 thymocytes, as well as by activated CD4+ and CD4+ CD45RO+ memory T cells (1). IL-17 stimulates the expression of TNFalpha, IL-1 beta, IL-6, and IL-8 (2). In addition, IL-17 is involved in the induction of inducible nitric oxide synthase and cyclooxygenase 2 in chondrocytes (3). High levels of IL-17 are associated with several chronic inflammatory diseases including rheumatoid arthritis, psoriasis and multiple sclerosis (4).

  • Q: I want to stain some T cell subsets (Th17 and Th1), do you have antibodies for that? I need for flow cytometer in sheep or goat.

    A:

    Unfortunately, we have neither a CD4 antibody nor an IL-17 antibody that has been validated in sheep or goat, although if you would like to try one you would again be eligible for our Innovators Reward Program.   • Q: I'm looking to establish an ELISA for IL-17 and want your recommendation on which antibody pairs to buy. Least expensive ones, please. Also, I'd like the detection antibody to be fluorescently labeled.

    Thank you for your patience while I reviewed our ELISA validated products for IL17. I was not sure what species you were going to be investigating, so I looked up our data for the human protein. If this is incorrect, just let me know and I can look up products for the species you will be working with. As for antibody pairs, we only had one set that have been tested together: Capture: NBP2-22531 Detection: NBP2-22532. The detection product is only available currently with a biotin conjugate. We can however offer custom conjugation for this product to any of our available conjugates. Please see the this link to see the available fluors. The typical fee for custom conjugation is usually about $80 additional. Once you know which fluor you would like, I can provide you with a quote. We do also offer several ELISA kits for this target (they typically employ HRP or Biotin detection systems, so I am not sure if they will work for your needs).
