Background: SHARPIN

Sharpin (SHANk-associated RH domain interacting protein) is a 387-amino acid protein, enriched in the postsynaptic density (PSD) of excitatory neurotransmitters. It is abundantly expressed in brain, spleen, lung, heart, skeletal muscle, kidney and testis (Stieglitz, 2012). A component of the large multi-protein E3 ubiquitin ligase complex called LUBAC, Sharpin regulates cell survival and apoptosis, cytokine production, and development of lymphoid tissues by regulating TNF-alpha induced canonical NF-kappaB signaling. Sharpin binds to the LUBAC subunit HOIP, and stimulates the formation of linear ubiquitin chains in vitro and in vivo. Sharpin harbors multiple functional motifs including an amino terminal coiled-coil (CC) domain, which has been shown to mediate the interaction between Sharpin and the scaffold protein Shank. The other two domains, UBL and NPL4 zinc finger domain facilitate ubiquitin-mediated protein recognition and degradation. Sharpin also acts as an inhibitor of beta1-integrins activation and down-regulates the activity of tumor suppressor protein PTEN, thus modulating immune and inflammatory diseases (Wang, 2012).Sharpin antibody recognizes endogenous levels of total Sharpin, which is recently identified as a commonly up-regulated gene in many human cancers and is believed to have a role in tumor biogenesis. It may function in small GTPase-mediated signaling pathways that may transform the actin cytoskeleton, thereby inducing morphological changes needed for tumor metastasis (Jung, 2010). Thus, Sharpin antibody plays an important role in normal immune development and control of inflammation.
