Specificity

This antibody is specific to 45-60 kD proteins identified as tau proteins. This antibody does not cross react with tubulin or other microtubule associated proteins. This antibody recognizes a phosphorylation-independent epitope in amino acids 404-441 (human). Tau antibody stains human neurofibrillary tangles, neutrophil threads and neurotic plaques associated with Alzheimer's disease.

Background: Tau

Principal Names: Microtubule-associated protein tau Official Gene Symbol: MAPT Gene ID: 4137 (Human) Gene Map Locus: 17q21.1 (Human). Tau is a low molecular weight component of cytoskeletal structures, normally functions as a microtubule associated protein, presumably by stabilizing the microtubules which serve as tracks for cytoplasmic transport. Alternative mRNA splicing of the the single tau gene on the long arm of human chromosome 17 gives rise to six brain tau proteins that are normally expressed in the adult human central nervous system. Inclusion of a 31 amino acid repeat encoded by exon 10 of the tau gene gives rise to the three isoforms with four microtubule-binding repeats each. The other three isoforms have three repeats each. Tau is mainly a neuronal protein, although its presence in different types of glia cells has been also reported in some neural diseases. Phosphorylation of tau by several phosphatases and kinases like GSK3 and Cdk5 affects its interaction with microtubules, resulting in the destabilisation of microtubules, ultimately leading to the degeneration of the affected neurons. Indeed hyperphosphorylated tau is the essential component of the different aberrant aggregates found in neurons (and sometimes in glia) of patients with neurological disorder like Alzheimers Disease where hyperphosphorylated tau is the major component of paired helical filaments (PHF) and thus neurofibrillary lesions. Filamentous tau protein deposits are also the defining characteristic of otherneurodegenerative diseases, many of which are frontotemporal dementias or movement disorders, such as Pick's disease, progressive supranuclear palsy, and corticobasal degeneration.
