Immunoprecipitation:ADAR AntibodyNBP3-29751] -Detection of human ADAR1 by western blot of immunoprecipitates. Samples: Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from HEK293T cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-ADAR1 antibody NBP3-29751 (lot NBP3-29751-3) used for IP at 3 µg per reaction. ADAR1 was also immunoprecipitated by rabbit anti-ADAR1.. For blotting immunoprecipitated ADAR1, NBP3-29751 was used at 0.4 µg/ml. Detection: Chemiluminescence with an exposure time of 30 seconds.

Background: ADAR

ADAR encodes the enzyme responsible for RNA editing by site-specific deamination of adenosines. This enzyme destabilizes double stranded RNA through conversion of adenosine to inosine. Mutations in this gene have been associated with dyschromatosis symmetrica hereditaria. Alternate transcriptional splice variants, encoding different isoforms, have been characterized.
