Calmodulin kinase II (CaMKII) is an 8 - 12 subunit multimeric calcium/calmodulin dependent Ser/Thr protein kinase. It is ubiquitously expressed and interacts with a very diverse group of substrates. In rat, there are four possible subunits/isozymes (alpha, beta, gamma, delta), each of which contains a catalytic, autoregulatory, and subunit-association domain.

Specificity
Specific for endogenous levels of the ~50 kDa alpha CaM Kinase II and the ~60 kDa beta CaM Kinase II proteins phosphorylated at Thr306. Immunolabeling is completely eliminated by treatment with lambda-phosphatase. It has been reported that this antibody may recognize some level of non-phosphorylated pure recombinant protein, but in our hands (as shown in our WB image) in native tissue, the antibody reacts in a phospho-specific manner.
