Western Blot: CBL Antibody [NBP1-28758] - Samples: Whole cell lysate from HeLa (5, 15 and 50 ug for WB; 1 mg for IP, 20% of IP loaded) and 293T (T; 50 ug) cells. Antibodies: Affinity purified rabbit anti-c-Cbl antibody NBP1-28758 used for WB at 0.04 ug/ml (A) and 0.4 ug/ml (B) and used for IP at 3 ug/mg lysate. c-Cbl was also immunoprecipitated by rabbit anti-c-Cbl antibody NBP1-28757, which recognizes an upstream epitope. Detection: Chemiluminescence with exposure times of 10 seconds (A) and 3 seconds (B).

Background: CBL

The cbl oncogene was first identified as part of a transforming retrovirus which induces mouse pre-B and pro-B cell lymphomas. As an adaptor protein for receptor protein-tyrosine kinases, it positively regulates receptor protein-tyrosine kinase ubiquitination in a manner dependent upon its variant SH2 and RING finger domains. Ubiquitination of receptor protein-tyrosine kinases terminates signaling by marking active receptors for degradation.
