This antibody was raised by immunizing mice with a B-Cell line derived from a Burkitt lymphoma.

Specificity

1.3.3.22 antibody recognizes CD81. Human CD81 is a member of the tetraspanin family of leukocyte cell surface molecules which are involved in signal transduction, cell-cell adhesion and cellular activation. CD81 also acts as a receptor for the envelope protein E2 of chronic hepatitis C virus. It is present on a wide variety of leukocytes.

The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom NBP3-12080, DAPI, merged channels and an isotype control. The isotype control was an unknown specificity antibody followed by staining with Alexa Fluor(R) 488 secondary antibody.

Flow Cytometry: CD81 Antibody (1.3.3.22) - Chimeric - Azide and BSA Free [NBP3-12080] - Paraformaldehyde fixed Daudi cells were stained with anti-unknown specificity antibody (isotype control, black line) or NBP3-12080 (blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.

Background: CD81

CD81/TAPA-1 is an integral membrane protein expressed on a variety of cell types, and has a high degree of sequence homology between human and mouse. (1) CD81 is expressed on thymic stromal cells where it plays an important role in the transition of gamma delta + T cells to more mature T cells with alpha beta T cell receptors. (2) Immunohistochemical staining has revealed that its expression is localized to the subcapsular region of the thymus and, specifically, on cells that have distinct clustering patterns. It has been speculated that the ligand for CD81 is the pre-T cell receptor, which is composed of a TCR beta chain and glycoprotein pT alpha. (2) The monoclonal antibody 2F7 can block thymocyte interaction with CD81 in vitro. (2)
