Description

This antibody is highly specific and optimized for use in Immunohistochemistry.

It has been validated on TMAs covering 76 normal tissues and numerous cancer tissues.

Positive Control
Postive control:Skin: All squamous epithelial cells (except the basal cell layer) should show strong cytoplasmic staining, while the basal cell layer remains KRT10 negative or shows only minimal staining.

Negative Control
Negitve control:Colon: All epithelial, inflammatory, stromal and muscle cells must not show any KRT10 staining.

Application Notes
Manual Protocol: Freshly cut sections should be used (less than 10 days between cutting and staining). Heat-induced antigen retrieval for 5 minutes in an autoclave at 121C in pH 7,8 Target Retrieval Solution buffer. Apply the antibody at a dilution of 1:150 at 37C for 60 minutes. Visualization of bound antibody by the EnVision Kit (Dako, Agilent) according to the manufacturer's directions.

Background: Cytokeratin 10

This gene encodes a member of the type I (acidic) cytokeratin family, which belongs to the superfamily of intermediate filament (IF) proteins. Keratins are heteropolymeric structural proteins which form the intermediate filament. These filaments, along with actin microfilaments and microtubules, compose the cytoskeleton of epithelial cells. Mutations in this gene are associated with epidermolytic hyperkeratosis. This gene is located within a cluster of keratin family members on chromosome 17q21. [provided by RefSeq, Jul 2008]
