The original antibody was generated by immunizing female BALB/c mice with an immunogenic polypeptide containing amino acids 410-432 (TIVEEVQDGKVISSREQVHQTTR) of human cytokeratin 17 conjugated with KLH. (Uniprot# Q04695)
Specificity
This antibody is specific for amino acids 410-432 (TIVEEVQDGKVISSREQVHQTTR) of human cytokeratin 17. Cytokeratins are intermediate filaments found in the intracytoplasmic cytoskeleton of epithelial tissue. Cytokeratin 17 regulates the activity of kinases such as PKC and SRC via binding to integrin beta-1 (ITB1) and the receptor of activated protein C kinase 1 (RACK1).

Flow Cytometry: Cytokeratin 17 Antibody (4D2) - Chimeric - BSA Free [NBP3-43565] - Paraformaldehyde fixed HeLa cells permeabilized with 0.5% Triton were stained with the anti-unknown specificity antibody (isotype control, black line) or NBP3-43565 (blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor® 488 antibody at a dilution of 1:1000, and the cells were analyzed using a FACSCanto flow-cytometer.Western Blot: Cytokeratin 17 Antibody (4D2) - Chimeric - BSA Free [NBP3-43565] -
Western Blot: Cytokeratin 17 Antibody (4D2) - Chimeric - BSA Free [NBP3-43565] - A431 (A) (0.001µg/ml) and HeLa (B) (0.0001µg/ml) cells lysate (35µg protein in RIPA buffer) were resolved on an SDS-PAGE gel, and blots were probed with NBP3-43565 at the mentioned respective concentrations before detection using an anti-rabbit secondary antibody. Immunocytochemistry/ Immunofluorescence: Cytokeratin 17 Antibody (4D2) - Chimeric - BSA Free [NBP3-43565] - Immunofluorescence analysis of paraformaldehyde fixed HeLa cells, permeabilized with 0.15% Triton, on coverslips stained with NBP3-43565 (1:100 dilution) for 1h followed by Alexa Fluor® 488 secondary antibody (1:1000 dilution), showing cytoplasmic staining. The nuclear stain is DAPI (blue). Panels show, from left-right, top-bottom,NBP3-43565, DAPI, merged channels, and an isotype control.
