Antibodies: Affinity purified rabbit anti-E6AP antibody used for WB at 0.4 mcg/ml (A) and 1 mcg/ml (B) and used for IP at 3 mcg/mg lysate. E6AP was also immunoprecipitated by rabbit anti-E6AP antibody, which recognizes an upstream epitope. Detection: Chemiluminescence with exposure times of 3 minutes (A and B).

Background: E6AP/UBE3A

E6AP was originally identified as the ubiquitin-protein ligase involved in human papillomavirus E6-mediated p53 degradation. It has since been shown to act as an E3 ubiquitin-protein ligase catalyzing the ubiquitination of several other protein substrates. Ubiquitin E3 ligases are overexpressed in many human cancers and their inhibition leads to growth suppression or apoptosis.
