Product Specifications

Immunogen

This affinity purified Fibroblast Activation Protein alpha/FAP Antibody was prepared from whole rabbit serum produced by repeated immunizations with a synthetic peptide corresponding to an internal region of mouse Fibroblast Activation Protein alpha/Fibroblast Activation Protein alpha/FAP protein. (Uniprot: P97321)

Reactivity Notes

A BLAST analysis was used to suggest partial cross-reactivity with Fibroblast Activation Protein alpha/FAP from human and rat sources based on ~88% homology with the immunizing sequence. Reactivity with Fibroblast Activation Protein alpha/FAP from other sources has not been determined. Human reactivity reported from a verified customer review.

Western Blot: Fibroblast Activation Protein alpha/FAP Antibody [NB110-85534]

Western Blot: Fibroblast Activation Protein alpha/FAP Antibody [NB110-85534]

Western Blot: Fibroblast Activation Protein alpha/FAP Antibody [NB110-85534] - Western blot using affinity purified anti-Fibroblast Activation protein alpha/Fibroblast Activation Protein alpha/FAP antibody shows detection of Fibroblast Activation Protein alpha/FAP protein in whole cell lysates from Fibroblast Activation Protein alpha/FAP expressing HEK cells (lane 4) but not control HEK cells (lane 3). Specific band staining is blocked when the primary antibody is pre-incubated with immunizing peptide (lanes 1 and 2 respectively). The band at ~90 kDa, indicated by the arrowhead, corresponds to the expected molecular weight of transfected Fibroblast Activation Protein alpha/FAP. Primary antibody was used at 1:1,000.
ELISA: Fibroblast Activation Protein alpha/FAP Antibody [NB110-85534]

ELISA: Fibroblast Activation Protein alpha/FAP Antibody [NB110-85534]

ELISA: Fibroblast Activation Protein alpha/FAP Antibody [NB110-85534] - ELISA results of purified Fibroblast Activation protein alpha/FAP antibody tested against BSA-conjugated peptide of immunizing peptide. Each well was coated in duplicate with 0.1ug of conjugate. The starting dilution of antibody was 5ug/ml and the X-axis represents the Log10 of a 3-fold dilution. This titration is a 4-parameter curve fit where the IC50 is defined as the titer of the antibody. Assay performed using 3% fish gel, Goat anti-Rabbit IgG Antibody Peroxidase Conjugated (Min X Bv Ch Gt GP Ham Hs Hu Ms Rt & Sh Serum Proteins) and TMB ELISA Peroxidase Substrate.

Identification of rAd‐infected DCs in vitro. (A–C) rAd‐FAP, rAd‐hlivin alpha, and rAd‐FAP/hlivin alpha were constructed using mouse FAP cDNA and human livin alpha cDNA, and then transduced into mouse bone marrow‐derived DCs, respectively. For evaluation of transduction efficacy, western blot was performed to measure protein expressions of FAP and livin alpha in cells, with GAPDH functioned as a loading control. ***p <.001, versus 1#; +++p <.001, versus 2#; ^^^p <.001, versus 3#. DCs, dendritic cells; FAP, fibroblast activation protein‐ alpha ; rAd‐FAP, recombinant adenoviral vector encoding mouse FAP cDNA; rAd‐hlivin alpha, recombinant adenoviral vector encoding human livin alpha cDNA. Background: Fibroblast Activation Protein alpha/FAP

Fibroblast Activation Protein (FAP) is expressed in the stroma of sites that are undergoing wound healing. In addition, it has recently been reported that FAP is expressed in the stroma of sites of metastatic disease. Inhibition of FAP may lead to a dramatic decrease in the number of metastatic osteosarcoma lung nodules. FAP exists as an inactive monomer and when activated forms homodimers or heterodimers with DPP4. Multiple isoforms of FAP are reported as alternative splicing products from a common gene.
