Background: Galectin-1

The galectins are a family of beta-galactoside-binding proteins implicated in modulating cell-cell and cell-matrix interactions. Galectin 1 may act as an autocrine negative growth factor that regulates cell proliferation. It also may regulate cell apoptosis and cell differentiation. It binds beta-galactoside, CD45, CD3 and CD4. Galectin 1 inhibits CD45 protein phosphatase activity and therefore the dephosphorylation of Lyn kinase. It is found in cardiac, smooth, and skeletal muscle, neurons, thymus, kidney, placenta and hematopoietic cells.

  • Q: Would you please tell me how mouse heart sample was prepared for the WB image from NBP1-95445?

    A: The protocol for tissue samples preparation for the WB with Galectin 1 antibody (# NBP1-95445 ) is as follows: 1. Wash the tissue with cold PBS. 2. Chop the tissue into small pieces, tip tissue sample into liquid nitrogen. 3. Start crushing tissue sample with pestle. Crushing sample to a powder before the liquid nitrogen completely evaporates. 4. Add appropriate volume of 1X hot lysis buffer, 100C, 5min. 5. Sonicate the tissue for 10-15seconds. 6. Spin tissue debris down at 10000rpm, 10min. 7. Carefully remove the lipid on the surface of the supernatant. Save supernatant as whole tissue lysate, and discard the pellet. 8. Determine the protein concentration by BCA protein assay. 9. Adjust concentration to 2 mg/ml with Lysis buffer. 10. Add an equal volume of 2X Laemmli Sample Buffer (final concentration of 1 mg/ml). 11. Boil each cell lysate in sample buffer at 100 C for 5 min and aliquot.
