Background: HNF-3 beta/FoxA2

This gene encodes a member of the forkhead class of DNA-binding proteins. These hepatocyte nuclear factors are transcriptional activators for liver-specific transcripts such as albumin and transthyretin, and they also interact with chromatin. Similar family members in mice have roles in the regulation of metabolism and in the differentiation of the pancreas and liver. This gene has been linked to sporadic cases of maturity-onset diabetes of the young. Two transcript variants encoding the same protein have been identified for this gene.

  • Q: I want to use the FoxA2 antibody (H00003170-M12) for Immunohistochemistry-Paraffin. Therefore I'm wondering if you can tell me which protocol you used to generate your picture? Which dilution did you take and which antigen retrieval method did you use in this case?

    A:

    Here is the link to the protocol that Abnova (the company that produces this antibody) uses on their site listed under the product (http://www.abnova.com/protocol_pdf/Immunohistochemistry.pdf). The dilution is 0.5 ug/mL. I would recommend citrate buffer pH 6.0 as the antigen retrieval as that is the one our lab has a lot of success with. We use a pressure cooker for the citrate buffer antigen retrieval method.
