Background: HP1 alpha

CBX5 (chromobox protein homolog 5), also known as HP1 (heterochromatin protein 1 homolog alpha), is one of 3 isoforms of HP1 that binds heterochromatin and functions as a structural adaptor to stabilize and organize macromolecular complexes with nucleosomes for higher chromatin structure. All of the HP1 isoforms possess a chromodomain and chromoshadow domain and associate with methylated histones that mediate transcriptional repression. Alternate names for CBX5 include HP1 alpha, antigen p25, HP1Hs-alpha, and HP1A.

  • Q: I want to use the hp1 alpha antibody in zebrafish, and I think NB110-40623 might work. The online information of this product mentions that it recognize the region between residue 150 and the C-terminus (residue 191) of human Chromobox Homolog 5. Here is my question, do you use this 42 amino acid peptide as immunogen? If not, can you tell me the exact sequence of immunogen, or its identidy to zebrafish hp1 alpha whose access no. is AAI64295.1.

    A: The immunogen for NB110-40623 was a synthetic peptide corresponding to the region 150-191, but the exact sequence can not be provided due to proprietary reasons. I did compare the homology between the human protein and the sequence you gave me, and it's greater than 95%.
