Background: SMURF2
SMURF2 (SMAD ubiquitin regulatory factor-2) is a ~86 kDa, 748 amino acid (aa) cytoplasmic E3 ubiquitin ligase that negatively regulates TGF-beta signaling by promoting proteasomal degradation of some SMAD signaling molecules. It also influences cell cycle progression, peaking during mitosis and binding MAD2L1 and NEDD9/CASL. SMURF2 contains C2, WW1-3, and HECT domains. A potential 735 aa form lacks aa 19-31 (within the C2 domain), while a 507 aa form shows an alternate start site at aa 242 (between WW1 and WW2). Within the HECT domain region used as an immunogen, human SMURF2 shares 99% and 98% aa identity with mouse and rat SMURF2, respectively.
