Western Blot: ICAM-1/CD54 Antibody (J0-F7) [NBP2-66896]

Western Blot: ICAM-1/CD54 Antibody (J0-F7) [NBP2-66896] - Western blot analysis of ICAM-1/CD54 on Daudi cell lysates. Proteins were transferred to a PVDF membrane and blocked with 5% BSA in PBS for 1 hour at room temperature. The primary antibody (1/500) was used in 5% BSA at room temperature for 2 hours. Goat Anti-Mouse IgG - HRP Secondary Antibody (HA1006) at 1:5,000 dilution was used for 1 hour at room temperature.

Formalin fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 10 minutes at room temperature and blocked with 1% Blocker BSA for 15 minutes at room temperature. Cells were probed with the primary antibody (M1511-6, 1/50) for 1 hour at room temperature, washed with PBS. Alexa Fluor(R))488 Goat anti-Mouse IgG was used as the secondary antibody at 1/1,000 dilution. The nuclear counter stain is DAPI (blue).

Flow Cytometry: ICAM-1/CD54 Antibody (J0-F7) [NBP2-66896] - Flow cytometric analysis of ICAM-1 was done on Jurkat cells. The cells were fixed, permeabilized and stained with the primary antibody (M1511-6, 1/100) (red). After incubation of the primary antibody at room temperature for an hour, the cells were stained with a FITC-conjugated Goat anti-Mouse IgG Secondary antibody at 1/200 dilution for 30 minutes.Unlabelled sample was used as a control (cells without incubation with primary antibody; black).

Background: ICAM-1/CD54

ICAM1 is a 85-110 kDa single chain type 1 integral membrane glycoprotein with an extracellular domain of five immunoglobulin superfamily repeats, a transmembrane region and a cytoplasmic domain. It shares considerable amino acid sequence homology with ICAM3 and with ICAM2. ICAM1 is expressed by activated endothelial cells. It is detected on cells of many other lineages (e.g. epithelial cells, fibroblasts, chondrocytes, B lymphocytes, T lymphocytes (low), monocytes, macrophages, dendritic cells and neutrophils), with lower levels that increase in inflammation. ICAM1 is also detected in some carcinoma and melanoma cells. Soluble ICAM1 is detectable in the plasma and is elevated in patients with various inflammatory syndromes. It is the receptor for rhinoviruses and malaria.
